摘要: 与正常小鼠外周血的红细胞不同,Lewis癌细胞(Lewis lung carcinoma, LLC)的荷瘤雄性小鼠的红细胞在外周血白细胞分离实验中并没有沉底,而是处于中间层。为探讨Sdr9c7基因的功能及荷瘤状态下红细胞发生异常的可能机制,采集实验小鼠的外周血,分离了红细胞、白细胞,并以Sdr9c7基因高表达的皮肤、肝脏、骨髓、血管作为对照组织,采用实时荧光定量PCR法检测Sdr9c7基因在正常小鼠和荷瘤小鼠各组织细胞中的表达变化情况。结果表明:相对于对照组,Sdr9c7基因唯独在荷瘤小鼠组的红细胞中表达显著上升,是正常小鼠红细胞的2.60±0.44倍;在皮肤、血管中表达无显著差异;在荷瘤小鼠组的血液、骨髓、肝脏、白细胞中的表达则显著下调,分别是正常小鼠相应组织细胞的0.31±0.05、0.44±0.04、0.36±0.04、0.35±0.08倍,表明荷瘤小鼠红细胞异常的发生可能与Sdr9c7基因表达上调有关。实验结果为进一步研究Sdr9c7基因的生物学功能拓宽视野,并为荷瘤状态下红细胞发生异常的机制提供新视角。
