摘要: 细菌耐药性已成为全球公共卫生的重大挑战,霍乱弧菌Vibrio cholerae作为一种主要的致病菌,其耐药性问题日益严重。本文基于全球霍乱弧菌基因组序列数据,利用生物信息学方法对其耐药基因(ARGs)的分布特征、丰度、多样性以及可移动性进行系统分析。通过对81 132条霍乱弧菌基因组序列的筛选与比对,共鉴定出16 570条ARG序列,涵盖83个ARG亚型,涉及19种抗生素类别。其中,多重耐药、四环素类、大环内酯类和磺胺类等ARGs丰度较高,占总体ARG丰度的69.23%。ARGs的多样性与丰度呈现显著地理异质性,中国、印度、海地等中低收入国家的水平显著高于高收入国家。高达91.57%的ARGs亚型与可移动遗传元件(MGEs)相关,凸显了MGEs在ARGs积累与传播中的关键作用。进一步通过聚类分析识别出120个可移动ARGs操作分类单元,其中26.67%在至少3个国家中检出,氨基糖苷类、氯霉素类、磺胺类等ARGs在全球范围内呈现出广泛的地理分布。代表性基因dfrA1、APH(6)-Id和cat3已证实发生洲际扩散,主要集中于亚非地区。本文研究为深入理解霍乱弧菌耐药性特征及其传播机制提供了科学依据,有助于制定有效的监测与防控策略,以应对霍乱弧菌及其他病原体的耐药性挑战。
