岩黄连总碱诱导肝星状细胞凋亡和自噬的电镜实验研究

Electron Microscopy Study on the Apoptosis and Autophagy of the Hepatic Stellate Cells Induced by Total Alkaloids

摘要: 为了研究岩黄连总碱诱导肝星状细胞凋亡和自噬的作用,本文通过体外培养HSC-T6细胞,应用MTT法检测不同浓度(0.20、0.25、0.30 g·L-1)脱氢卡维丁组、巴马汀组和小檗碱组的细胞增殖抑制率,应用透射电子显微镜观察各组HSC-T6细胞的超微结构改变,结果发现:3组不同药物诱导的HSC-T6细胞抑制率均呈浓度依赖性;经不同浓度3组药物处理24 h后的HSC-T6细胞均呈典型的细胞凋亡超微结构改变,并出现不同程度的自噬现象。可见,脱氢卡维丁、巴马汀和小檗碱能诱导HSC-T6细胞凋亡,也能诱导细胞自噬。这3种成分可能为岩黄连总碱中抗肝纤维化的有效成分。

Abstract: To study the effects of total alkaloids on apoptosis and autophagy of hepatic stellate cells in vitro.Cell inhibition rates which using different concentrations (0.20、0.25、0.30 g·L-1) of Dehydrocavidine group, Pakistan Martin group and Berberine group to the cultured HSC-T6 cells in vitro were detected.The ultrastructural changes of each group cells were observed by transmission electron microscopy. The results showed that, the cell inhibition rates of HSC-T6 cells induced by different drugs in three groups were dependent on concentrations;the HSC-T6 cells showed typical ultrastructural changes of apoptosis after 24h’s processing with different concentrations of the drugs,and there were different levels of the autophagy.Cavitin, Martin and berberine could induce apoptosis and autophagy of the HSC-T6 cells. There three components may be effective to anti liver fibrosis in total alkaloids of the Coptis chinensis Franch.
