南方红豆杉提取物的抗氧化、抗肿瘤活性研究

Antioxidant and Antitumor Activities of Extracts from Taxus chinensis var. mairei

摘要: 在超声波辅助和80 ℃下,以无水乙醇为溶剂对南方红豆杉进行提取,浓缩后得提取物EE,EE通过柱色谱法分离制得紫杉醇,并对EE和紫杉醇的抗氧化、抗肿瘤活性进行研究。结果表明,EE对DPPH·、ABTS+·、O-2·、OH· 的清除活性显著优于常用抗氧化剂BHT和紫杉醇,半清除浓度IC50分别为13.19±1.50、2.65±0.35、25.33±1.75和343.00±2.13 mg/L,说明EE具有优良的抗氧化活性;紫杉醇对人非小肺癌细胞A549、人乳腺癌细胞MCF-7、人肝癌细胞Bel-7402和子宫颈癌细胞Hela的抑制活性远优于EE,半抑制浓度IC50分别为3.45±1.37、6.20±0.88、1.98±0.49和1.15±0.27 mg/L。

Abstract: Under ultrasound, Taxus chinensis var. mairei were extracted with ethanol at 80 ℃ to offer ethanol extract (EE). EE was further extracted with chloroform and separated by column chromatography to offer taxol. Their antioxidant and antitumor activities were then evaluated. The antioxidant activity screening results showed that EE exhibited better scavenging activity than the common antioxidant BHT in these four assays, with IC50 of 13.19±1.50, 2.65±0.35, 25.33±1.75 and 343.00±2.13 mg/L, respectively, while taxol showed the worst, indicating excellent antioxidant activity of EE. The antitumor activity screening results demonstrated that taxol showed better inhibition than EE, with IC50 of 3.45±1.37, 6.20±0.88, 1.98±0.49 and 1.15±0.27 mg/L, respectively.
