The Role of DDX17 RNA Helicase in SET8 Response to Wnt Pathway

摘要: Wnt信号通路是一个庞大的网络体系,由许多转录因子和调节蛋白参与到该通路的调节过程中,提供了该通路的细胞类型、发育时空等的精细调节。本实验通过实时定量PCR和Co-IP等实验,探讨RNA解旋酶DDX17在SET8参与的Wnt信号通路调控中的作用。结果表明,DDX17抑制Wnt靶基因的表达;DDX17与转录抑制因子HDAC1、SET8和LEF1相互作用。

Abstract: Wnt signaling is composed by a huge network system in which many transcription factors and regulatory proteins are involved in the pathway.The role of the DDX17 RNA helicase in the SET8 response to Wnt signaling by RT-PCR and Co-IP methods is studied.The result preliminary finds that DDX17 is crucial for activation of the Wnt target genes in mammalian cells.Additionally,there is interaction between DDX17s and SET8/TCF4/LEF1/HDAC1.
