Effect of Endurance Exercise on HO-1 Expression and Its Resistance to Non-Alcoholic Fatty Liver Disease Formation

Abstract: To explore the relationship of moderate intensity endurance exercise on Heme Oxygenase-1 (HO-1) protein expression in liver and its resistance to Non-Alcoholic Fatty Liver Disease Formation of rats fed with high-fat diet, 48 male SD rats were randomly divided into normal diet control group (NC), exercise and normal diet group (EN), the inhibitor + exercise and normal diet group (DEN) and high fat diet control group (HC), exercise and high fat diet group (EH), the inhibitor + exercise and high fat diet group (DEH). The changes of blood TC, TG, FFC concentrations and the expression & the activity of HO-1 protein in the liver were observed, as well as for the changes of Total Antioxidant Capacity & MDA content and the changes of blood ALT, AST concentrations & liver morphology. The results showed that, in the groups of NC, EN and DEN, the expression and activity of HO-1 protein & the Total Antioxidant Capacity of liver in group EN were the highest. Compared with group NC, groups HC, EH and DEH showed significantly higher concentrations of blood TC, TG, FFC, and a much higher expression of HO-1 protein with a significantly decreased HO-1 activity and a Total Antioxidant Capacity in the liver, as well as much higher MDA content in the liver and serum ALT, AST levels which suggested typical liver fatty degeneration. Among them, the liver HO-1 protein expression of group EH was the highest, but the above-mentioned abnormal changes for group EH were the lightest. In addition, the expression of HO-1 protein in the liver of group EH was significantly higher than that of group EN. Some conclusions could be drawn from these studies. High fat diet easily induced dyslipidemia in rats, decreased HO-1 activity and antioxidant capacity of liver, and increased reactive oxygen species. Moderate intensity endurance exercise can induce the expression of HO-1 protein in liver, increase the HO-1 activity in the livers of rats fed with high fat diet and the ability of antioxidation to eliminate reactive oxygen species, preventing NAFLD formation and protecting liver function.
